This protocol uses readily available tools to safely and quickly sample endosperm tissue from seeds of maize or other plants with relatively large seeds. Several common DNA extraction methods are also evaluated for use with the chipped tissue. We prefer the SDS method for the overall economy, or the filter plate method when DNA quality is the main priority.
A peer-reviewd manuscript describing the protocol has been published in bio-protocol (Zebosi et al. 2025, DOI: 10.21769/BioProtoc.5200).
A video to accompany the protocol is in the publication and permanently available at Figshare.
